Capillary electrophoresis is used to quantify the mtDNA amplified in the PCR process. Each PCR-amplified sample is run with a 200-bp standard of known concentration. The peak for the 200-bp standard is shown on the left in this electropherogram, and the peak for the PCR-amplified DNA is on the right. The area under the 200-bp standard peak is compared to the area under the peak of the unknown sample to estimate the concentration of DNA present in the sample. This estimate is used to prepare the cycle sequencing reactions.
